ProQR rebuilt around its Axiomer RNA-editing platform after exiting ophthalmology, and AX-0810's Phase 1 data in healthy volunteers showing an 8-fold bile acid increase — exceeding a predefined 2-fold threshold — delivered the platform's first clinical proof of concept. The thesis resolves on whether that pharmacodynamic signal translates to histologic or clinical benefit in pediatric biliary atresia patients, where the Phase 2 interim readout is the make-or-break event. The principal approval-path risk is that the biliary atresia program relies on an investigator-initiated trial in China before a potentially registration-enabling study, a geography and design combination that could complicate FDA and EMA data acceptance.
