SION-719 wipeout forces a pivot to an unvalidated dual combo; AscenSION CF POC readout is the only remaining test that matters

Sionna is a clinical-stage company developing a targeted therapy to improve cystic fibrosis lung function beyond existing approved treatments, though its lead candidate is not yet approved.

Sionna was built to improve CFTR function beyond Trikafta in cystic fibrosis via a novel NBD1-stabilization mechanism, but PreciSION CF delivered a -1.0 mmol/L placebo-adjusted sweat chloride change, a clear null result, and SION-719 was discontinued. What remains is a speculative pivot: SION-451 paired with galicaftor in the AscenSION CF trial, backed by Phase 1 safety and PK data only, with no efficacy signal in CF patients. The pivot faces structurally long odds because the same sweat chloride endpoint that defeated SION-719 will adjudicate SION-451/SION-2222, and the post hoc rationale offered for the prior failure has not been prospectively validated.
